Description: CA77.1 is an activator of chaperone-mediated autophagy (CMA). CA77.1 activates chaperone-mediated autophagy (CMA) by increasing the expression of the lysosomal receptor for this pathway, LAMP2A, in lysosomes. CA77.1 is a derivative of AR7. CA77.1 may help to degrade toxic pathogenic protein products such as tau proteins and has potential applications in the treatment of Alzheimer's disease.

Biological target: CA77.1 can increase the expression of the lysosomal receptor LAMP2A in lysosomes. CA77.1 improves behavior and neuropathology in a mouse model with systemic and neuronal-specific CMA blockage.
In vitro activity: To be determined
In vivo activity: The chemical enhancement of CMA ameliorates pathology in two different Alzheimer's disease experimental mouse models. CM77.1 activates CMA. This study concludes that functional CMA is essential for neuronal proteostasis through the maintenance of a subset of the proteome with a higher risk of misfolding than the general proteome.
