MedKoo product information:
SNS-314
Description of SNS-314: SNS-314 is a synthetic
small molecule Aurora kinase (AK) inhibitor with potential
antineoplastic activity. Aurora kinase inhibitor SNS-314 selectively
binds to and inhibits AKs A and B, which may result in the
inhibition of cellular division and proliferation in tumor cells
that overexpress AKs. AKs are serine-threonine kinases that play
essential roles in mitotic checkpoint control during mitosis. Check
for
active clinical trials or
closed clinical trials using this agent. (NCI

Information about this agent
|
SNS-314 is a potent and sele ctive inhibitor of Aurora kinases A, B, and C. Proliferating cells treated with SNS-314 bypass the
mitotic spindle checkpoint and fail to undergo cytokinesis, leading to
multiple rounds of endoreduplication and eventually cell death.
NS-314 is currently in a dose escalation phase 1
clinical trial for the treatment of patients with advanced solid tumors.
Both the in vitro synergies observed between SNS-314 and agents that
target the mitotic spindle and the potentiation seen with docetaxel in
vivo are consistent with a mechanism of action in which Aurora
inhibition bypasses the mitotic spindle assembly checkpoint and prevents
cytokinesis, augmenting subsequent spindle toxin-mediated mitotic
catastrophe and cell death. For detail see:
